Stains live animal cells with green fluorescence and dead cells with red fluorescence for detection by fluorescence microscopy or flow cytometry

• Calcein-AM is a cell-permeable, non-fluorescent ester that can passively enter cells. In viable cells, it is hydrolysed by esterases in the cytoplasm to release the green fluorescent dye calcein. Calcein itself is impermeable to membranes and is only retained in viable cells with an intact plasma membrane
• Ethidium homodimer III is a red fluorescent nucleic acid dye that is impermeable to membranes. It selectively colours dead cells with damaged plasma membranes (necrotic or late apoptotic cells)
Membrane impermeable nucleic acid dyes can be used to selectively stain late necrotic and apoptotic cells with leaky membranes
