GSTrap columns are designed for one-step purification of GST-tagged proteins. The columns are prepacked with the matrices:
• Glutathion Sepharose HP: for high-resolution purifications, from a more concentrated sample;
• Glutathion Sepharose FF: for high flow rate purifications;
• Glutathion Sepharose 4B, standard support.
Available in 2 formats: 1 or 5 ml column.
GST fusion protein cleavage enzyme
Used for site-specific separation of the GST tag from proteins:
• expressed using pGEX-T vectors: Thrombin Protease;
• expressed using pGEX-X vectors: Facto Xa Protease;
• expressed using pGEX-6P vectors: PreScission Protease.
PreScission Protease: one unit will cleave >90 % of 100 µg of a test GST-fusion protein in 50 mm Tris-HCl, 150 mm NaCl, 1 mm EDTA, 1 mm DTT, pH 7.0 at 5 °C for 16 h.
Thrombin Protease: one unit of enzyme cleaves 90% of 100 ?g of a test GST fusion protein when incubated in 1 × PBS at 22 °C for 16 h
Factor Xa Protease: one unit cleaves >90 % of 100 ?g of a test GST fusion protein when incubated in 1 mm CaCl?, 100 mm NaCl, and 50 mm Tris-HCl (pH 8.0) at 22 °C for 16 h.
